I got a 1981 Yamaha xs400, and my rectifier is dead. I'm not picking up anything on my multimeter with it.

Does this explain why I have no spark?

How did you check the rectifier? It will only output DC when AC is applied to it, so in other words the motor needs to be turning. You can check the rectifier when it's unplugged for continuity, it should only pass in one direction. Most multimeters have a function for this, some better ones have a dedicated diode function as well.

so how do I test it when I'm turning the motor over?
The rectifier, along with the voltage regulator, is only for charging the battery. You won't need it to produce a spark, however your battery will die pretty quick without it.
If the engine is running and your seeing 14ish volts DC on your battery then your rectifier is good. That is normal charging voltage.
To get a spark all you need is the pickup coil, tci, charged battery and ignition coils. When I was having charging problems I just used my truck battery jumpered to the bike (truck running).
